摘要:
目的:通过观察Survivin拮抗肽对C6胶质瘤细胞生长抑制作用,探究Survivin拮抗肽对C6胶质瘤细胞增殖的作用机制.方法:1.MTT法检测Survivin拮抗肽对C6大鼠胶质瘤细胞增殖抑制影响.2. 应用免疫组化法检测Survivin拮抗肽作用前后实验组和对照组C6大鼠胶质瘤细胞中Caspace-3的表达情况.结果:MTT结果显示随着Survivin拮抗肽浓度的增加,对C6胶质瘤细胞的增殖抑制作用逐渐增强,且随浓度及作用时间延长抑制率上升,20?g/mlSurvivin拮抗肽作用72h后,抑制率为62%,呈剂量依赖性,免疫组化显示随着Survivin拮抗肽浓度的增加,Caspace-3在C6大鼠胶质瘤细胞细胞核的表达逐步增加.结论:Survivin拮抗肽可使caspase-3 在细胞核中的表达增多,从而促进细胞凋亡,可能对C6 胶质瘤细胞发挥较强的抑制作用。
Abstract:
Objective: To explore the mechanism of Survivin antagonistic peptide on the proliferation of C6 glioma cells through investigating the inhibitory effect of Survivin antagonistic peptide on the proliferation of C6 glioma cells. Methods: MTT assay was used to detect the inhibitory effect of Survivin antagonist peptide on the proliferation of C6 rat glioma cells .Immunohistochemical method was used to detect the expression changes of Caspace-3 in experimental and control groups with or without Survivin antagonistic peptide in C6 rat glioma cells. Results: MTT results showed that with the inhibited effect of C6 glioma cells proliferation was strengthen in compare with the concentration increase of Survivin antagonistic peptide. What is more, the inhibitory rate rised in a dose- and time- dependent manner.Treatment with 20 ?g/ml Survivin antagonist peptide for 72h, the inhibitory rate was 62%, which was exhibited in a dose-dependent manner. Immunohistochemistry showed that with the increase of Survivin antagonistic peptide concentration, further the expression of Caspace-3 in the nuclei of C6 rat glioma cells was gradually increased. Conclusion: Survivin antagonistic peptide could up-regulate the expression of caspase-3 expression in the nucleus, further promoting cell apoptosis, and may play a strong inhibitory effect on C6 glioma cells.
