摘要:
目的:探讨miR-221/miR-222在胶质瘤患者脑脊液中的表达及其诊断价值。方法:采用实时荧光定量PCR检测31例胶质瘤患者和33例非胶质瘤患者的脑脊液miR-221/miR-222表达水平,并采用ROC曲线评价其诊断胶质瘤的价值。结果:miR-221和miR-222在胶质瘤患者脑脊液中的表达水平均明显高于对照组(均P<0.001)。高级别胶质瘤患者的脑脊液miR-221、miR-222表达水平高于低级别组,其中miR-222表达水平的差异具有统计学意义(1.49倍,P=0.007)。脑脊液miR-221、miR-222的表达水平都能将胶质瘤患者与对照者区分开;而二者联合诊断胶质瘤的曲线下面积(AUC)为0.849,灵敏度为81.8%,特异度为83.9%。结论:脑脊液中高表达的miR-221/miR-222有可能作为胶质瘤诊断的生物标志物。
Abstract:
Objective To explore different expression of miR-221/miR-222 in cerebrospinal fluid (CSF) between glioma patients and controls,and investigate the potential role of CSF miR-221/miR-222 in detecting glioma. Methods A total of 31 glioma patients and 33 nonglioma patients (controls) were enrolled in our study. Expression levels of CSF miR-221/miR-222 were analyzed by quantitative reverse transcription polymerase chain reaction (qRTPCR).Diagnostic value of CSF miR-221/miR-222 was assessed by receiver operating characteristic (ROC) method. Results CSF miR-221 and miR-222 were significantly higher in glioma patients compared with controls (all P<0.001).The expression of miR-221/miR-222 in the CSF of patients with high grade glioma was higher than that in low grade glioma,and the difference of miR-222 expression was statistically significant (1.49 fold,P=0.007).The area under the ROC curve (AUC) of the combination of the two miRNAs was 0.849,with sensitivity of 81.8% and specificity of 83.9%. Conclusion miR-221/miR-222 in CSF can serve as a biomarker in diagnosing glioma.
