摘要:
目的 研究脑胶质瘤组织中Hippo通路核心分子Yap,Taz在蛋白水平的表达情况及其亚细胞定位,分析它们与脑胶质瘤的相关性。方法 采用免疫印迹(Western Blot,WB)及免疫组化(Immunohistochemical,IHC)技术检测Hippo通路相关蛋白Yap、Taz在非肿瘤脑组织及脑胶质瘤组织中的表达情况。结果 WB检测发现Yap和Taz在胶质瘤组织中表达水平较非肿瘤脑组织高,其表达水平随肿瘤级别增高而增高, 差别有统计学意义(*P<0.05,**P<0.01);免疫组化检测发现Yap 和 Taz 均主要定位于细胞核中,在胶质瘤组织中表达水平较非肿瘤脑组织高,其表达水平随肿瘤级别增高而增高,差别有统计学意义(**P<0.01)。结论 Yap和Taz在人脑胶质瘤组织中表达水平升高,其表达水平随胶质瘤级别增高而增高。
Abstract:
Objective To study the protein level, subcellular localization of the hippo pathway core molecules Yap and Taz in human gliomas. Methods The protein levels of Yap and Taz in the nontumorous brain tissues and glioma samples were examined by Western Blot and Immunohistochemical staining. Result The levels of Yap and Taz were up-regulated in glioma tissues examined by Western Blot. The protein levels of Yap and Taz increased with the glioma grade increase, The difference was statistically significant (*P<0.05,**P < 0.01). IHC showed that both Yap and Taz were mainly located in nucleus, The levels of Yap and Taz were up-regulated in glioma tissues examined by Western Blot. The protein levels of Yap and Taz increased with the glioma grade increase, The difference was statistically significant. Conclusion Yap and Taz are highly expressed in human brain glioma samples and the higher the glioma grade, the higher the levels of Yap and Taz.
