摘要:
目的探讨不同级别脑胶质瘤及瘤旁组织中长链非编码RNA CRNDE的差异表达,以及CRNDE的表达水平与肿瘤临床、病理特征之间的相关性。方法采集2007年—2017年在河北省儿童医院神经外科行脑胶质瘤切除术获取的肿瘤组织及对应的瘤旁组织标本164例。通过实时荧光定量PCR(quantitative realtime PCR)技术分别检测CRNDE的相对表达水平,并比较其在不同级别脑胶质瘤及瘤旁组织中的表达水平。分析164例胶质瘤组织CRNDE表达水平与临床、病理特征之间的关系。采用KaplanMeier法进行单因素分析,并绘制生存曲线。CRNDE表达水平对患者生存时间的影响采用Logrank法分析。结果与瘤旁组织(1.00±0.06)比较,脑胶质瘤组织的CRNDE表达水平(3.49±1.71)显著上调,差异有统计学意义(P<0.05)。与低级别胶质瘤组(1.51±0.86)相比,高级别胶质瘤组的CRNDE表达水平(4.26±1.29)升高更显著,差异有统计学意义(P<0.05)。脑胶质瘤组织的CRNDE表达水平与患者的性别、年龄、术前症状、肿瘤是否坏死之间均无相关性(均P>0.05),而与肿瘤的直径、病理级别、复发之间有相关性(列联系数分别为0.2,0.38,0.2;P=0.01-0.001)。CRNDE高表达组和低表达组患者1年生存率分别为91.4%及 96.1%(P<0.05);高表达组和低表达组5年生存率分别为23.2%及59.1%(P<0.05)。结论CRNDE在人脑胶质瘤组织的表达水平较瘤旁组织显著升高;其表达水平与胶质瘤病理级别有关;高级别脑胶质瘤的表达水平高于低级别脑胶质瘤。CRNDE的表达水平与胶质瘤的病理级别、肿瘤直径及肿瘤复发之间有正相关性,而与患者的性别、年龄等因素无相关性。CRNDE高表达患者的生存时间短。
Abstract:
Abstract:ObjectiveTo investigate the expression of CRNDE in glioma of different pathological grades and adjacent non-tumor tissues, and to explore the association between CRNDE expression levels and clinical, pathological characteristics. MethodsA total of 164 tumor samples and adjacent non-tumor tissues were collected from patients who underwent surgical resection in the Department of Neurosurgery, Hebei province Children’s Hospital from 2007 to 2017. Quantitative real-time PCR(qRT-PCR) were performed to detect CRNDE expression levels in adjacent non-tumor tissues and glioma tissues of different grades for comparison. Chi-square test were used to investigate the relationship between clinicopathological characteristics and CRNDE mRNA expression levels in 164 glioma tissues. Univariate analysis were performed by the Kaplan-Meier method and survival curves were constructed. The effects of CRNDE expression on survival time were investigated and analyzed using Log-rank analysis. ResultsCompared with adjacent non-tumor tissues(1.00±0.06), expression level of CRNDE in glioma tissues(3.49±1.71) was highly up-regulated, and the differences were statistically significant(P<0.05). CRNDE expression level in high-grade gliomas(Ⅲ—Ⅳ) (4.26±1.29) was significantly higher than that in low-grade gliomas(Ⅰ—Ⅱ) (1.51±0.86), showing significant difference(P<0.05). There was no correlation between CRNDE expression level and gender, age, preoperative symptoms as well as tumor necrosis, CRNDE mRNA expression level was correlated with tumor size, pathological grades of tumor and tumor recurrence. The contingency coefficients between CRNDE mRNA expression level and tumor size, tumor grade as well as tumor recurrence were 0.2, 0.38 and 0.2 respectively. The 1 year survival rate in high and low CRNDE expression group were 91.4% and 96.1%(P<0.05). The 5 year survival rate in high and low CRNDE expression group were 23.2% and 59.1%(P<0.05). ConclusionsExpression levels of CRNDE mRNA were significantly higher in human glioma tissues than in adjacent nontumor tissues, which were associated with pathological grades of glioma. CRNDE expression levels in highgrade gliomas(Ⅲ—Ⅳ) were higher than those in lowgrade gliomas(Ⅰ—Ⅱ), suggesting significant difference. CRNDE expression level was positively correlated with WHO grading, tumor size, and tumor recurrence, other than gender and age. Overall survival time was shorter in those with high expression of CRNDE.
