【摘要】目的 研究下调磷酸甘油酸激酶1(PGK1)对U373细胞的放疗增敏作用;探讨PGK1参与的放疗增敏的可能机制。方法建立放射抵抗U373(RR-U373)细胞;采用LipofectamineTM 2000将shRNA-PGK1转染至U373细胞;对照组为0 Gy,处理组为5 Gy放射治疗。应用MTT法、划痕实验、Matrigel侵袭实验分别检测放疗前后各组细胞的增殖能力、迁移能力和侵袭能力;Western-Blot法检测各组细胞PGK1、CIN(chronophin)和丝切蛋白1(cofilin 1,CFL1)蛋白表达量。结果在U373细胞和RR-U373细胞中,相对于对照组,下调PGK1的细胞在放疗前后的增殖、迁移及侵袭能力均减弱;下调PGK1表达后,CIN、CFL1蛋白表达水平降低。结论下调U373细胞中PGK1 的表达对胶质瘤U373细胞有放疗增敏作用。PGK1可能通过调节CIN、CFL1蛋白表达影响胶质瘤细胞对放射的敏感性。
Abstract:
Abstract: ObjectiveTo explore the role of down-regulating phosphokinase 1 (PGK1) in ameliorating the radiosensitivity of U373 human glioma cell and the potential mechanism. MethodsRadioresistant U373 cells (RR-U373) were established. The short hairpinRNA (shRNA) for PGK1 were transfected to RR-U373 cells and normal U373 cells using LipofectamineTM 2000. After transfection, they were treated with 5 Gy or 0 Gy irradiation. MTT assay, scratch assay and transwell assay were used to determine the relative survival, cell migration and cell invasion abilities, respectively. Western Blot was uesd to evaluate PGK1,CIN and CFL1 expressions. ResultsDownregulating PGK1 using shRNA decreased cell viability,migration and invasion abilities both before and after radiation.After down-regulation of PGK1, the expression of CIN and CFL1 protein decreased. ConclusionThese findings suggest that down-regulating PGK1 could enhance radiosensitivity in U373 human glioma cell and the efficacy may be associated with the regulation effect of PGK1 on CIN and CFL1.
