目的研究原肌球蛋白1(tropomyosin 1,TPM1)与胶质瘤U251细胞放射抵抗性之间的关系,为进一步寻找提高胶质瘤的放射敏感性的可能靶点提供理论依据。方法转染shRNA-TPM1(pcDNA3.1-TPM1)抑制(诱导)U251和RR-U251细胞的TPM1表达。各组细胞进行放射治疗后,采用MTT法、划痕实验、侵袭实验、流式细胞术分别检测U251(RR-U251)细胞的增殖、迁移、侵袭、凋亡能力。用Western Blot检测TPM1蛋白表达水平,进而观察各组细胞放射敏感性的变化。结果与U251细胞相比,RR-U251细胞的TPM1蛋白表达水平明显下降(P<0.01)。各组细胞进行放射治疗后,与对照组相比,沉默TPM1的RR-U251和U251细胞的增殖、迁移和侵袭能力均显著提高,细胞凋亡率降低(均P<0.05)。TPM1基因过表达时逆转了这一现象。结论TPM1可提高胶质瘤U251细胞的放射敏感性。TPM1与胶质瘤细胞的放射抵抗机制有关,其可能是改善胶质瘤放射敏感性的潜在靶点。
Abstract:
Objective To study the relationship between tropomyosin TPM1 and radiosensitivity of glioma U251 cells and to provide a basis for further research on possible targets for improving the radiosensitivity of glioma. Methods TPM1 levels in the U251 and RR-U251 cells were inhibited by transfection with shRNA-TPM1 while overexpression was induced by treatment with pcDNA3.1-TPM1. After radiation treatment of cells in each group, MTT assay, wound-healing assay, transwell chamber method, flow cytometry were used to determine the cell proliferation ability, migration ability, invasion ability, apoptosis ability of various U251(RR-U251)cells, respectively. Western Blot was used to observe changes in the protein and radiosensitivity of each group of cells. Results Compared with U251 cells, the expression level of TPM1 was signifcantly decreased in the RR-U251 cells(P<0.05). After radiation treatment of cells in each group, the proliferation, migration and invasion ability of RR-U251 and U251 cells silenced by TPM1 were notably improved and the apoptosis rate was reduced compared with the control group(all P<0.05). This phenomenon was reversed by TPM1 gene overexpression. Conclusions TPM1 can increase the radiosensitivity of glioma U251 cells. TPM1 is related to the mechanism underlying glioma radiation resistance and may be a potential target for improving the radiosensitivity of glioma.
