摘要:
胶质瘤是中枢神经系统最常见、致死率最高的恶性肿瘤。与其他恶性肿瘤一样,胶质瘤起源于细胞的基因突变。研究表明,带有基因突变的细胞要生长为一个具有全部表型的恶性肿瘤,需经过一个有细胞增殖、分化、干性和细胞间相互作用参与的异常发育过程。在这一过程中,肿瘤细胞“征用”了大量的正常发育调控信号通路。Notch信号通路是介导相邻细胞间相互作用的,在进化中高度保守的信号通路,广泛参与正常发育的精确调控和恶性肿瘤的发生发展过程。本文对Notch信号通路及其在胶质瘤中的作用和机制进行综述,并探讨以Notch信号通路为靶点干预和治疗胶质瘤的前景。
Abstract:
Gliomas are the most common malignancy with highest mortality in the central nervous system. Like other malignancies, gliomas originate from genetic mutations in cells. However, studies have shown that cells with genetic mutations need to undergo an abnormal developmental process involving cell proliferation, differentiation, stemness and cell-cell interaction in order to grow into a tumor with full malignant phenotypes. In this process, tumor cells “enlist” a large number of normal developmental regulatory signaling pathways. Notch signaling pathway is a highly conserved signaling pathway that mediates the interaction between adjacent cells and is widely involved in the development of normal tissues and malignant tumors. This article will review the progress of the role and mechanisms of Notch signaling pathway in gliomas, and discuss the prospect of using Notch signaling pathway as a target for intervention and treatment of gliomas.
